Adanın sahəsi 41 214 km² təşkil edir. Adaların dünyada sahəsinə görə sıralamasında 34-cü yerdə gedir. Sahil xətdinin uzunluğu 1944 km təşkil edir.
Sauthempton adası Melvill yarımadasından Frozen-Streyt boğazı vastəsi ilə ayrılır. Hudzon körfəzinin qərb sahilindən isə onu Ros-Velyam boğazı ayırır. Ən hündür nöqtəsi Matiasen dağıdır. Bell yarımadası cənub-şərqdə yerləşir.
Sauthempton adası Kanada ərazisində qış vaxtına keçməyən az saylı ərazilərdəndir.
